I had invested $500 on Google AdWords call campaign and Got ROI $250.
Is there any way to make this campaign profitable?

Yes. If you can get the half of what you've spent on first run, it looks you can make it profitable. You have to spend some time optimizing and keep adding negative keywords that don't convert. To be honest Plumber is not cheap at all on AdWords and you should very wisely choose the keywords you bid on. I will advise you not to bid on the keywords that everyone in this niche is using in their campaigns. If still expensive and not profitable, switch to another niche.
